I received a new USB Drive today courtesy of Slickdeals and since it's 4x larger than my current one, I went a bit wild looking for programs and cool things to do. A few cool things:
PStart - Small program that can be set to autorun when you insert the USB drive. Basically, it's an advanced toolbar. You can add shortcuts and folders from your USB drive for easy access.
PortableApps (http://portableapps.com/) - Cool site that has a lot of applications that you can install on a USB drive to run from any computer and still keep the data (browser bookmarks, chat logs and such).
Acronis - Total suite for recovery & backup. There's also a program that lets you copy files to your USB drive so you can boot with a special (linux?) OS that you can use to clone, backup, recover or read data from your hard drives.
